Title
How will indel characteristics change in different cell types after DNA double stranded breaks?
Abstract
The understanding behind how stem cells and their differentiated lines repair DNA double stranded breaks (DSBs) are largely unknown. This project repurposes a lineage tracing system as a genetic barcode and an inducible Cas9 platform to induce and track the indels in human induced pluripotent stem cells and neural progenitors. By comparing indel outcomes across the cell types, we can infer DSB repair pathways differentially recruited in each cell type. This provides insight into how repair factor activity varies with cell identity and can contribute to broader efforts of providing solutions for genetic diseases such as Fanconi Anemia.
